普魯士藍(lán)納米粒子的光熱毒性研究

【摘要】:在科學(xué)技術(shù)高速發(fā)展的今天,學(xué)科交叉已經(jīng)成為一大趨勢,其中納米材料在醫(yī)學(xué)領(lǐng)域的應(yīng)用尤為矚目。納米材料因其獨特的生物醫(yī)學(xué)優(yōu)勢有望解決傳統(tǒng)醫(yī)學(xué)方法的生物醫(yī)學(xué)問題,引領(lǐng)現(xiàn)代醫(yī)學(xué)診斷和治療方法的新變革。然而當(dāng)前多數(shù)納米材料的研究不夠深入,其在人體內(nèi)的生物安全性得不到保障,且很多材料制備復(fù)雜、價格昂貴,嚴(yán)重限制了其進(jìn)一步發(fā)展。因此,納米毒理學(xué)應(yīng)運而生。它與傳統(tǒng)毒理學(xué)相互滲透并逐漸發(fā)展,融入納米技術(shù)、材料科學(xué)、生物學(xué)、醫(yī)學(xué)、毒理學(xué)、藥理學(xué)等方面的基礎(chǔ)知識,對納米技術(shù)的安全性以及人類社會至關(guān)重要。同時,納米材料光熱治療日漸興起,經(jīng)過激光照射后,利用納米光熱劑表面等離子體共振產(chǎn)熱,產(chǎn)生高溫殺死腫瘤細(xì)胞,可以達(dá)到治療癌癥的目的。普魯士藍(lán)是一種古老的試劑,其制備簡單、價格便宜、光熱性能優(yōu)異。最為重要的是普魯士藍(lán)長期的臨床用藥驗證了其在人體內(nèi)可靠的生物安全性和代謝途徑。正是由于這些優(yōu)勢,普魯士藍(lán)的納米材料一旦被深入應(yīng)用于醫(yī)學(xué)研究,將有潛力解決其它多數(shù)納米材料生物安全性得不到保障且價格昂貴的醫(yī)學(xué)難題,其研究和應(yīng)用價值巨大。作為一種優(yōu)良光熱試劑因其良好的光熱性能和極高的生物相容性被用于生物醫(yī)學(xué)領(lǐng)域,然而其光熱致死癌細(xì)胞的毒性機(jī)理卻鮮有人問津。本論文針對普魯士藍(lán)納米粒子(PB NPs)的光熱性質(zhì)、光熱穩(wěn)定性、細(xì)胞毒性、細(xì)胞光熱毒性進(jìn)行了探究,分析了對比了四種細(xì)胞He La、Chang liver、4T1、L929在不同濃度PB納米粒子濃度和不同激光功率密度下的細(xì)胞死亡方式,并進(jìn)一步探討了影響這些細(xì)胞光熱產(chǎn)生致死性的毒性的因素。
[Abstract]:With the rapid development of science and technology, interdisciplinary has become a major trend, among which the application of nano-materials in the field of medicine is particularly attractive.Nanomaterials, due to their unique biomedical advantages, are expected to solve the biomedical problems of traditional medical methods and lead to new changes in modern medical diagnosis and treatment.However, most of the current research on nanomaterials is not deep enough, their biological safety in human body can not be guaranteed, and many materials are complex and expensive, which seriously limits their further development.Therefore, nanometer toxicology arises at the historic moment.It permeates with traditional toxicology and develops gradually. It is very important for the safety of nanotechnology and human society to integrate the basic knowledge of nanotechnology, material science, biology, medicine, toxicology, pharmacology and so on.At the same time, photothermal therapy of nanomaterials is emerging day by day. After laser irradiation, the surface plasmon resonance (SPR) of nano-photothermal agent is used to produce heat and kill tumor cells at high temperature, which can be used to treat cancer.Prussian blue is an ancient reagent, it is simple to prepare, cheap, excellent photothermal properties.The most important thing is that Prussian blue has proven its biological safety and metabolic pathway in human body.Because of these advantages, Prussian Blue nanomaterials, once used in medical research, will have the potential to solve the other nanomaterials biological safety can not be guaranteed and expensive medical problems, its research and application is of great value.As a kind of excellent photothermal reagent, it has been used in biomedical field because of its good photothermal properties and high biocompatibility. However, the toxic mechanism of photothermal lethal cancer cells is rarely studied.In this paper, the photothermal properties, photothermal stability, cytotoxicity and photothermal toxicity of Prussian blue nanoparticles (PB NPs) were investigated.In this paper, the cell death patterns of four kinds of He-La-Chang Chang livertio 4T _ (1) L _ (929) cells under different concentrations of PB nanoparticles and different laser power densities were analyzed and compared, and the factors influencing the lethal toxicity of these cells were discussed.
