三種不同銀納米粒子SERS基底比較研究

【摘要】:表面增強(qiáng)拉曼光譜(Surface-Enhanced Raman Spectra,簡稱SERSp)技術(shù)是一種新興的分析檢測技術(shù),由于其對樣品分析時,具有靈敏度高、檢測時間短以及樣品所需量小等優(yōu)點(diǎn),在最近幾年里,SERSp技術(shù)已在眾多的領(lǐng)域如:生物醫(yī)學(xué),藥物學(xué),化學(xué)等都得到了廣泛的應(yīng)用,同時表面增強(qiáng)拉曼散射(Surface-Enhanced Raman Scattering,簡稱SERS)基底的制備也已成為該領(lǐng)域的研究熱點(diǎn)。本文不僅介紹了拉曼光譜相關(guān)的理論知識,而且重點(diǎn)對基于銀納米粒子的三種SERS基底進(jìn)行了研究比較。本文的主要內(nèi)容如下:首先,本文從拉曼散射的基本原理、拉曼光譜的基本特征以及拉曼光譜在生物醫(yī)學(xué)及藥物學(xué)中的應(yīng)用與前景這幾方面對拉曼光譜進(jìn)行了概述。其次,對表面增強(qiáng)拉曼光譜進(jìn)一步論述,介紹了表面增強(qiáng)拉曼光譜的發(fā)現(xiàn)與發(fā)展,同時重點(diǎn)介紹了表面增強(qiáng)拉曼散射的兩種增強(qiáng)機(jī)理:基于電磁場效應(yīng)的增強(qiáng)機(jī)理和基于化學(xué)效應(yīng)的增強(qiáng)機(jī)理。最后,重點(diǎn)對三種以銀納米粒子(AgNPs)的SERS效應(yīng)為基質(zhì)的拉曼活性基底:毛細(xì)管-AgNPs基底,二氧化鈦-AgNPs基底和濾紙-AgNPs基底進(jìn)行了比較研究。分別用這三種基底對羅丹明6G(R6G)分子進(jìn)行拉曼光譜數(shù)據(jù)的采集及分析,從而找出這三種SERS基底相應(yīng)的最佳制備條件。最后用這三種最佳條件下制備的SERS基底對同一個健康人血清進(jìn)行拉曼光譜檢測,并對結(jié)果進(jìn)行分析比較。初步結(jié)果:這三種基于AgNPs的SERS基底都是可靠的和實(shí)用的;二氧化鈦-AgNPs基底靈敏度相對較高,但制備過程較復(fù)雜;濾紙-AgNPs基底靈敏度其次;毛細(xì)管-AgNPs基底及濾紙AgNPs基底的制備均較為簡單。因此,從實(shí)用的角度考慮,濾紙-AgNPs基底比較適合血清的表面增強(qiáng)拉曼光譜的檢測與分析。
[Abstract]:Surface-enhanced Raman Spectra (SERSpS) technology is a new analytical and detection technique, which has the advantages of high sensitivity, short detection time and small amount of sample. In recent years, SERSp technology has been widely used in many fields such as biomedicine, pharmacology, chemistry and so on. At the same time, the preparation of surface-enhanced Raman scattering (SERS-based) substrates has become a hotspot in this field. Three kinds of SERS substrates based on silver nanoparticles are studied and compared. The main contents of this paper are as follows: firstly, the basic principle of Raman scattering is introduced. The basic characteristics of Raman spectrum, the application and prospect of Raman spectrum in biomedicine and pharmacology are summarized. Secondly, surface enhanced Raman spectroscopy is further discussed. In this paper, the discovery and development of surface-enhanced Raman spectroscopy are introduced, and two enhancement mechanisms of surface-enhanced Raman scattering are emphasized: the enhancement mechanism based on electromagnetic field effect and the enhancement mechanism based on chemical effect. Three kinds of Raman active substrates based on the SERS effect of silver nanoparticles were focused on: capillary-AgNPs substrates. A comparative study of titanium dioxide-AgNPs substrate and filter paper-AgNPs substrate was carried out. The Raman spectra data of Rhodamine 6GN R6G) molecules were collected and analyzed by using these three substrates. The optimal preparation conditions of the three SERS substrates were found out. Finally, the Raman spectra of the same healthy serum were detected by using the SERS substrates prepared under these three optimal conditions. The three kinds of SERS substrate based on AgNPs are all reliable and practical, the sensitivity of titania-AgNPs substrate is relatively high, but the preparation process is more complicated, the sensitivity of filter paper-AgNPs substrate is second; The preparation of capillary-AgNPs substrate and filter paper AgNPs substrate is relatively simple. Therefore, the filter paper-AgNPs substrate is more suitable for the detection and analysis of serum surface enhanced Raman spectroscopy from a practical point of view.
