【摘要】:目的:采用仿生硅化法實(shí)現(xiàn)對(duì)胰蛋白酶的固定,并將固定化酶與聚甲基丙烯酸甲酯(PMMA)相結(jié)合用于抗生物粘泥復(fù)合膜的制備。方法:研究了正硅酸甲酯(TMOS)與酶的濃度比、介孔氧化硅(MPS)與酶的質(zhì)量比、有機(jī)溶劑等因素的影響。并以MPS固定化酶為對(duì)照實(shí)驗(yàn),對(duì)復(fù)合膜酶的穩(wěn)定性、催化活性、重復(fù)使用性、抗蛋白質(zhì)吸附性能等進(jìn)行了分析。結(jié)果:通過仿生硅化法制備的仿生氧化硅-酶-聚甲基丙烯酸甲酯復(fù)合膜(Si O_2-Enzyme-PMMA film),與介孔氧化硅-酶-聚甲基丙烯酸甲酯復(fù)合膜(MPS-Enzyme-PMMA film)相比,具有更好的穩(wěn)定性,在25℃和50℃儲(chǔ)藏30 d后,兩種膜酶活性分別為86.3%和81.2%。Si O_2-Enzyme-PMMA film在重復(fù)使用6次之后,復(fù)合膜的酶活剩余89.1%。將復(fù)合膜浸泡在牛血清白蛋白(BSA)溶液中18 d后,其表面蛋白質(zhì)吸附量?jī)H為不含酶PMMA的三分之一。兩種復(fù)合膜均具有良好的抗蛋白吸附性能。結(jié)論:仿生硅化制備固定化酶,與介孔氧化硅載體相比,在提高固定化酶的包埋率和穩(wěn)定性方面有更顯著的優(yōu)勢(shì),其條件溫和,操作簡(jiǎn)便,活性高,應(yīng)用于抗生物粘泥涂層具有更好的防污效果。
[Abstract]:Objective: to immobilize trypsin by bionic silicification. The immobilized enzyme and polymethyl methacrylate (PMMA) were combined to prepare the composite membrane. Methods: the concentration ratio of methyl orthosilicate (TMOS) to polymethyl methacrylate (PMMA) was studied. The effect of mass ratio of mesoporous silica to enzyme, organic solvent and so on. The stability, catalytic activity and reusability of the composite membrane enzyme were studied by using MPS immobilized enzyme as control. Results: the bionic silica-enzyme-poly (methyl methacrylate) composite membranes were prepared by bionic silicification. SiO2-Enzyme-PMMA film.
