UPLC-Q-TOF-MS法測定小鼠大腦中4種單胺類神經(jīng)遞質(zhì)的含量

【摘要】:建立了超高效液相色譜-四極桿飛行時間質(zhì)譜法(UPLC-Q-TOF-MS)對小鼠大腦中的多巴胺(DA)、5-羥色氨(5-HT)、二羥基苯乙酸(DOPAC)和高香草酸(HVA)4種單胺類神經(jīng)遞質(zhì)的含量進行同時測定。樣品以30%乙醇溶液(甲酸調(diào)至p H 5.0)勻漿處理后,冷凍離心,過濾。在優(yōu)化色譜-質(zhì)譜條件下,采用ESI和APCI復(fù)合源,正/負離子掃描方式進行數(shù)據(jù)采集。4種單胺類神經(jīng)遞質(zhì)在0.5~1 000μg/L范圍內(nèi)呈良好線性關(guān)系,相關(guān)系數(shù)(r)大于0.999 3,方法的檢出限為0.5~5.0μg/L,平均回收率為93.3%~101.8%,相對標(biāo)準(zhǔn)偏差為1.1%~3.3%。該方法具有操作簡便、高效快速、回收率良好、靈敏度高、重現(xiàn)性好和準(zhǔn)確度高等優(yōu)點,可用于小鼠腦組織內(nèi)4種單胺類神經(jīng)遞質(zhì)的含量測定。
[Abstract]:An ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) was established for the treatment of dopamine (DA), 5-hydroxytryptamine (5-HT) in the brain of mice. The contents of four monoamine neurotransmitters, dihydroxyphenylacetic acid (DOPAC) and hyaluronic acid (HVA), were determined at the same time. The sample was homogenized with 30% ethanol solution (formic acid to pH 5.0), frozen and centrifuged and filtered. Under the condition of optimized chromatography-mass spectrometry, the data were collected by positive / negative ion scanning with ESI and APCI composite sources. Four monoamine neurotransmitters showed a good linear relationship in the range of 0.5 鈮,
