侵襲性感染金黃色葡萄球菌的毒力基因系統(tǒng)研究

【摘要】:目的研究侵襲性感染金黃色葡萄球菌分離株的毒力基因存在狀況,為臨床治療提供參考依據(jù)。方法收集杭州市余杭區(qū)中醫(yī)院2013年2-7月20株金黃色葡萄球菌侵襲性感染臨床分離株,采用聚合酶鏈反應(yīng)(PCR)的方法,分析共6大類94種毒力基因;并對毒力基因檢測結(jié)果作菌株系統(tǒng)發(fā)育分析,數(shù)據(jù)采用SPSS 17.0軟件進(jìn)行統(tǒng)計(jì)分析。結(jié)果 20株侵襲性感染金黃色葡萄球菌中共檢測到66種毒力基因:包括11種黏附毒素毒力基因、6種細(xì)胞毒素毒力基因、3種侵襲性酶毒力基因、12種超抗原毒力基因、32種外毒素毒力基因、2種莢膜抗原毒力基因;系統(tǒng)發(fā)育分析結(jié)果為20株金黃色葡萄球菌可分為A、B兩個(gè)簇群,其中B簇群中的11、14號株和2、3、19號株可能為克隆播散。結(jié)論侵襲性感染金黃色葡萄球菌攜帶多種毒力基因,具有極強(qiáng)的侵襲性和定植能力,對人體致病性強(qiáng),對金黃色葡萄球菌人體侵襲性感染分離株進(jìn)行黏附毒素、細(xì)胞毒素、侵襲性酶、超抗原、外毒素、莢膜抗原等六大類94種毒力基因檢測研究為國內(nèi)首次。
[Abstract]:Objective to study the existence of virulence genes in invasive Staphylococcus aureus isolates and to provide reference for clinical treatment. Methods from February to July 2013, 20 clinical isolates of invasive infection of Staphylococcus aureus were collected from Yuhang Hospital, Hangzhou City, and 94 virulence genes of 6 categories were analyzed by polymerase chain reaction (PCR). The results of virulence gene detection were analyzed by phylogenetic analysis. The data were statistically analyzed by SPSS 17.0 software. Results 66 virulence genes were detected in 20 strains of invasive Staphylococcus aureus, including 11 virulence genes of adhesion toxin, 6 virulence genes of cytotoxin, 3 virulence genes of invasiveness enzyme and 12 virulence genes of superantigen. 32 exotoxin virulence genes, 2 capsular antigen virulence genes; Phylogenetic analysis showed that 20 strains of Staphylococcus aureus could be divided into A and B clusters, of which 11, 14 and 2,3, 19 strains were probably cloned and disseminated. Conclusion Staphylococcus aureus with invasive infection has many virulence genes, it has strong invasiveness and colonization ability, and has strong pathogenicity to human body. It can be used as adhesion toxin and cytotoxin to human invasive infection strains of Staphylococcus aureus. Invasive enzyme, superantigen, exotoxin, capsule antigen and other 94 virulence genes were detected for the first time in China.
