【摘要】：目的研制一種解脲支原體(Uu)膠體金免疫層析試劑盒,用于臨床快速檢測、流行病學調查。方法原核表達解脲支原體MB(Uu-MB)蛋白,制備Uu-MB單抗后膠體金標記,采用雙抗體夾心法制備試劑盒,評價試劑盒的特異性、敏感性、穩(wěn)定性及符合率。結果對Uu陽性培養(yǎng)物和Uu-MB具有良好特異性;37℃放置12d質量穩(wěn)定;試劑盒與(Uu)支原體分離鑒別管的檢測陽性率分別為29.59%和71.42%;Uu金標檢測試劑盒與Uu分離鑒別管的陽性、陰性符合率為41.28%、100.0%;Uu分離鑒別管培養(yǎng)物經PCR鑒定,兩者的陽性、陰性符合率為100%、75.65%。結論研制的試劑盒操作簡便,具有特異性強、敏感性高等特點,是一種適宜普及推廣的Uu感染檢測產品。
[Abstract]:Objective to develop a gold immunochromatographic kit for rapid detection and epidemiological investigation of Ureaplasma Urealyticum (Uu) colloid. Methods Ureaplasma Urealyticum MB (Uu-MB) protein was expressed in E. coli and labeled with colloidal gold after preparation of Uu-MB monoclonal antibody. The kit was prepared by double antibody sandwich method. The specificity, sensitivity, stability and coincidence rate of the kit were evaluated.
