不同貯存時(shí)間的單采少白細(xì)胞血小板中精氨酸酶水平的研究

【摘要】：目的:探討不同貯存時(shí)間對(duì)單采少白細(xì)胞血小板(apheresis leukocyte-reduced platelet,ALR-Plt)中精氨酸酶水平的影響及精氨酸酶的可能來(lái)源。方法:應(yīng)用ELISA法檢測(cè)不同貯存時(shí)間ALR-Plt和對(duì)照血漿中精氨酸酶水平及髓過(guò)氧化物酶(myeloperoxidase,MPO)水平;應(yīng)用相關(guān)性分析分析不同貯存時(shí)間的ALR-Plt中精氨酸酶水平與MPO水平的相關(guān)性。結(jié)果:貯存3 d內(nèi)的ALR-Plt與對(duì)照血漿中精氨酸酶水平差異無(wú)統(tǒng)計(jì)學(xué)意義,但是貯存超過(guò)3d的ALR-Plt中精氨酸酶水平顯著高于貯存3 d內(nèi)的ALR-Plt和對(duì)照血漿中的水平(P0.05)。不同貯存時(shí)間的ALR-Plt之間MPO水平差異無(wú)統(tǒng)計(jì)學(xué)意義,但均顯著低于對(duì)照血漿中的水平(P0.05)。相關(guān)性分析顯示:ALRPlt中精氨酸酶水平與MPO水平呈正相關(guān)(r=0.58)。結(jié)論:貯存時(shí)間超過(guò)3 d的ALR-Plt中精氨酸酶水平顯著升高,其可能來(lái)源于殘留的少量白細(xì)胞,特別是中性粒細(xì)胞。
[Abstract]:Aim: to investigate the effect of different storage time on argininase level in single white blood cell platelets (apheresis leukocyte-reduced platelet,ALR-Plt) and the possible sources of argininase. Methods: the levels of arginase and myeloperoxidase (myeloperoxidase,MPO) in ALR-Plt and control plasma were detected by ELISA method, and the correlation between arginase level and MPO level in ALR-Plt with different storage time was analyzed. Results: there was no significant difference in arginase level between ALR-Plt and control plasma after 3 days of storage, but the arginase level in ALR-Plt stored for more than 3 days was significantly higher than that in ALR-Plt and control plasma (P0.05). There was no significant difference in MPO levels between ALR-Plt at different storage time, but they were significantly lower than those in control plasma (P0.05). Correlation analysis showed that argininase level was positively correlated with MPO level (r = 0. 58). Conclusion: the level of argininase in ALR-Plt for more than 3 days may be derived from a small amount of residual white blood cells, especially neutrophils.
