【摘要】：目的觀察血液樣本處理方式對大鼠血液生化指標檢測結果的影響。方法取體重180~220 g的SPF級Wistar大鼠40只,雌雄各半,禁食不禁水12 h后,用3%戊巴比妥鈉麻醉后取全血和肝素抗凝血各3 m L。將每種血樣分別置于3個1.5 m L的離心管和肝素抗凝管中各1 m L,并分別在室溫放置1 h、2 h、4 h后進行離心,測定血糖、血脂、蛋白、肝功能、腎功能、心肌酶等指標。結果血液樣本類型和采集后離心前的放置時間對Wistar大鼠的血清或血漿TC、TG、TP、ALB、ALT、ALP、BUN和CREA等指標均無明顯影響(P0.05)。但血漿的GLU值顯著高于血清(P0.05,P0.01),并隨著采集后離心前放置時間的延長持續(xù)下降。血清AST和CK、LDH值則均顯著高于血漿(P0.05,P0.01),且隨采集后離心前放置的延長呈現(xiàn)持續(xù)顯著的上升(P0.05,P0.01)。而采集后離心前放置時間對Wistar大鼠血漿AST和CK、LDH值均無明顯影響(P0.05)。結論血液樣本類型對血脂、蛋白和腎功能指標無明顯影響,但對GLU、AST、CK、LDH等檢測結果有一定影響。采集后離心前的放置時間延長可導致血清血漿GLU下降,血清AST、CK、LDH升高,但對血漿AST、CK、LDH指標無明顯影響。
[Abstract]:Objective to observe the effect of blood sample treatment on blood biochemical indexes in rats. Methods Forty SPF grade Wistar rats weighing 180 ~ 220g, male and female, were selected. After fasting for 12 hours, the whole blood and heparin anticoagulant blood were taken after anaesthesia with 3% pentobarbital sodium. Each blood sample was placed in 3 1.5 mL centrifuge tubes and 1 mL heparin anticoagulant tube respectively. The blood glucose, blood lipid, protein, liver function, renal function and myocardial enzyme were measured by centrifugation at room temperature for 1 h and 2 h for 4 h. Results the type of blood sample and the time before centrifugation had no significant effect on the serum or plasma TCCT TGV, alt, ALP, bun and CREA of Wistar rats. However, the GLU value of plasma was significantly higher than that of serum P0.05, P0.01C, and decreased with the prolongation of the time before centrifugation. The values of serum AST and CKD were significantly higher than that of plasma P0.05U P0.01g, and the values of serum AST and CKD showed a continuous and significant increase with the prolongation of pre-centrifugation after collection (P0.05U P0.01a). However, the time before centrifugation had no significant effect on the levels of plasma AST and CKD in Wistar rats. Conclusion Blood sample type has no significant effect on blood lipid, protein and renal function, but it has some influence on the detection results of GLU ASTX CKD and LDH. The prolongation of the storage time before centrifugation resulted in the decrease of plasma GLU and the increase of serum ASTCKN LDH, but there was no significant effect on the plasma ASTK GLU index.
