【摘要】:目的分析中國(guó)南方人群HPA-15基因型分布頻率,研究血小板上HPA-15抗原表達(dá)量。尋找較好的血小板保存方法以維持HPA-15的高表達(dá)量,為單克隆抗體俘獲血小板抗原技術(shù)(MAIPA)檢測(cè)HPA-15同種抗體提供合適的供者血小板。方法 1)隨機(jī)選取廣州血液中心的健康血小板單采獻(xiàn)血者109名,留取EDTA抗凝管全血。SSPPCR方法檢測(cè)HPA-15基因型。流式細(xì)胞術(shù)檢測(cè)血小板上HPA-15抗原表達(dá)量。2)另隨機(jī)選取健康獻(xiàn)血者的單采血小板67例,將每份血小板分為2份,1份重懸于EDTA-PBS中放于4℃保存作為檢測(cè)組A,另1份留于血小板辮子中放于血小板振蕩儀中作為檢測(cè)組B,在d 0、1、2、3、7、14檢測(cè)2組血小板上HPA-15的檢測(cè)含量,比較2組HPA-15含量的變化。結(jié)果 1)109名隨機(jī)健康獻(xiàn)血者HPA-15a和HPA-15b的基因頻率是0.495和0.505。流式細(xì)胞術(shù)檢測(cè)HPA-15抗原的MFI值的范圍2.44-20.4,陽性率3.2%-86.7%。不同HPA-15基因型之間,HPA-15抗原表達(dá)量有統(tǒng)計(jì)學(xué)差異(P0.05)。2)分析比較AB這2個(gè)組,隨著測(cè)量時(shí)間點(diǎn)的推移,2組HPA-15的表達(dá)量均有降低,但檢測(cè)組B中HPA-15的表達(dá)含量高于檢測(cè)組A,且2組的MFI值有統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論 HPA-15a和HPA-15b的基因分布頻率相近,HPA-15抗原表達(dá)含量整體水平偏低且個(gè)體差異性大。體外保存方式下,HPA-15的表達(dá)含量隨著時(shí)間推移逐漸降低,4℃保存方式下HPA-15含量降解尤為明顯。血小板振蕩儀中保存血小板的方法為MAIPA檢測(cè)HPA-15抗體提供更合適的血小板抗原。
[Abstract]:Objective to analyze the distribution frequency of HPA-15 genotypes and to study the expression of HPA-15 antigen on platelets in southern China. To find a better method of platelet preservation to maintain the high expression of HPA-15 and to provide suitable donor platelets for monoclonal antibody capture platelet antigen (MAIPA) detection of HPA-15 homoantibodies. Methods 1) A total of 109 healthy platelet donors from Guangzhou Blood Center were randomly selected for detection of HPA-15 genotypes by SSPPCR in the whole blood samples of EDTA anticoagulant tubes.
