B_w31的血型血清學(xué)和分子生物學(xué)研究——附1例報(bào)告

【摘要】:目的對(duì)1例正反定型不符的標(biāo)本進(jìn)行血清學(xué)疑難血型鑒定和分子生物學(xué)機(jī)制分析。方法應(yīng)用常規(guī)血型血清學(xué)方法進(jìn)行血型鑒定;對(duì)ABO基因的Exon6和Exon7擴(kuò)增后進(jìn)行直接測(cè)序,并進(jìn)行單體型測(cè)序確定其基因型。結(jié)果該例標(biāo)本血清學(xué)表現(xiàn)為B_w,紅細(xì)胞與抗-A不凝集,與抗-B 2+凝集,與抗-A1不凝集,血清中存在抗-A;直接測(cè)序及單體型測(cè)序顯示,此例標(biāo)本的B等位基因在Exon7存在nt664GA突變。結(jié)論經(jīng)測(cè)序檢測(cè)證實(shí),此例血清學(xué)表現(xiàn)為B_w的標(biāo)本,其基因型為罕見的B_w31/O01型。
[Abstract]:Objective to identify the difficult blood group and analyze the molecular biological mechanism of a sample with positive and negative typing. Methods routine blood group serological method was used to identify the blood group, the Exon6 and Exon7 of ABO gene were amplified and sequenced directly, and their genotypes were determined by haplotype sequencing. Results the serological manifestations of this sample were: B _ W, erythrocyte and anti--A agglutination, anti-B _ 2 agglutination, anti-A _ 1 agglutination, and serum anti--A.The direct sequencing and haplotype sequencing showed that the B allele of this sample had nt664GA mutation in Exon7. Conclusion the serological features of this serological specimen were confirmed by sequencing, and the genotype of BW was a rare B_w31/O01 type.
