【摘要】：目的對1例正反定型不符的標本進行血清學(xué)疑難血型鑒定和分子生物學(xué)機制分析。方法應(yīng)用常規(guī)血型血清學(xué)方法進行血型鑒定;對ABO基因的Exon6和Exon7擴增后進行直接測序,并進行單體型測序確定其基因型。結(jié)果該例標本血清學(xué)表現(xiàn)為B_w,紅細胞與抗-A不凝集,與抗-B 2+凝集,與抗-A1不凝集,血清中存在抗-A;直接測序及單體型測序顯示,此例標本的B等位基因在Exon7存在nt664GA突變。結(jié)論經(jīng)測序檢測證實,此例血清學(xué)表現(xiàn)為B_w的標本,其基因型為罕見的B_w31/O01型。
[Abstract]:Objective to identify the difficult blood group and analyze the molecular biological mechanism of a sample with positive and negative typing. Methods routine blood group serological method was used to identify the blood group, the Exon6 and Exon7 of ABO gene were amplified and sequenced directly, and their genotypes were determined by haplotype sequencing. Results the serological manifestations of this sample were: B _ W, erythrocyte and anti--A agglutination, anti-B _ 2 agglutination, anti-A _ 1 agglutination, and serum anti--A.The direct sequencing and haplotype sequencing showed that the B allele of this sample had nt664GA mutation in Exon7. Conclusion the serological features of this serological specimen were confirmed by sequencing, and the genotype of BW was a rare B_w31/O01 type.
