基于量子點(diǎn)熒光探針的高靈敏蛋白質(zhì)檢測(cè)方法

【摘要】:通過(guò)制備量子點(diǎn)熒光檢測(cè)探針,構(gòu)建了一種基于量子點(diǎn)探針和免疫磁珠的蛋白質(zhì)檢測(cè)方法,實(shí)現(xiàn)了對(duì)蛋白腫瘤標(biāo)志物癌胚抗原(carcinoembryonic antigen,CEA)的高靈敏定量檢測(cè).在該檢測(cè)體系中,若存在有靶標(biāo)蛋白,其與量子點(diǎn)檢測(cè)探針以及捕獲探針之間會(huì)發(fā)生免疫反應(yīng)形成三明治結(jié)構(gòu),利用磁力分離器對(duì)免疫復(fù)合物進(jìn)行富集后,通過(guò)檢測(cè)富集在磁珠表面的量子點(diǎn)熒光信號(hào),可實(shí)現(xiàn)對(duì)靶標(biāo)蛋白的定量.該方法的檢測(cè)靈敏度為38 pg/mL,線性范圍為0.39~50ng/mL,臨床質(zhì)控樣本檢驗(yàn)結(jié)果表明,該方法準(zhǔn)確度高,可重復(fù)性好,可應(yīng)用于臨床樣本檢測(cè).該量子點(diǎn)探針檢測(cè)體系具有靈敏度高、特異性好、樣品消耗量低等優(yōu)點(diǎn),在疾病早期診斷方面具有廣闊的應(yīng)用前景.
[Abstract]:A method of protein detection based on quantum dot probe and immunomagnetic beads was constructed by preparing quantum dot fluorescence detection probe. A highly sensitive and quantitative detection method for carcinoembryonic antigen (CEA), a protein tumor marker, was developed. If there is a target protein, the immune reaction between the target protein and the quantum dot detection probe and the capture probe will form a sandwich structure, and the immune complex can be enriched by magnetic separator. The quantification of target protein can be achieved by detecting the fluorescence signals of quantum dots enriched on the magnetic bead surface. The sensitivity of the method is 38 PG / mL and the linear range is 0.39 ~ 50 ng / mL. The results of clinical quality control samples show that the method is accurate and reproducible. The quantum dot probe detection system has the advantages of high sensitivity, good specificity and low sample consumption, so it has a broad application prospect in the early diagnosis of disease.
