【摘要】：目的研究溶血磷脂酶C(plcH)對(duì)銅綠假單胞菌肺部感染者肺功能的作用機(jī)制,為銅綠假單胞菌肺部感染的靶向治療提供參考。方法選擇8~12周齡成年雄性C57BI/6J小鼠共9只,隨機(jī)分為3組,即PAO1組、ΔplcH組和對(duì)照組(即PBS組),每組各3只,對(duì)9只小鼠進(jìn)行肺感染造模;分別在LB培養(yǎng)基中培養(yǎng)銅綠假單胞菌株P(guān)AO1(野生型,即WT型)和等基因ΔplcH缺失株用于小鼠肺感染造模;測(cè)定小鼠通氣與肺功能,分析小鼠肺泡表面活性物質(zhì)功能;采用SPSS 19.0軟件進(jìn)行數(shù)據(jù)統(tǒng)計(jì)分析。結(jié)果小鼠感染24h后,PAO1組和ΔplcH組的脾臟和肺臟內(nèi)的銅綠假單胞菌含量差異均無(wú)統(tǒng)計(jì)學(xué)意義;PBS組BALF中的白細(xì)胞計(jì)數(shù)顯著低于其他兩組(P0.05);P-V曲線和下降的滯后吸氣肢評(píng)估結(jié)果顯示,PAO1組出現(xiàn)極差的肺病理生理學(xué),與PBS組小鼠相比,ΔplcH組小鼠的肺部生理功能顯著異常,但好于PAO1組。結(jié)論銅綠假單胞菌plcH可能通過(guò)作用于肺表面活性物質(zhì)從而影響肺功能。
[Abstract]:Objective to study the mechanism of lysophospholipase (LPL) on pulmonary function of pulmonary infection infected by Pseudomonas aeruginosa (Pseudomonas aeruginosa). Methods Nine adult male C57BI / 6J mice at the age of 812 weeks were randomly divided into three groups: PAO1 group. Nine mice were induced by lung infection in 螖 plcH group and control group (PBS group, 3 rats in each group). Pseudomonas aeruginosa PAO1 (wild type, WT type) and isogenic 螖 plcH deletion strain were cultured in LB medium, respectively. The ventilatory and pulmonary function of mice were measured, and the function of alveolar surfactant in mice was analyzed. SPSS 19.0 software was used to analyze the data. Results the mice were infected for 24 hours. There was no significant difference in the content of Pseudomonas aeruginosa in spleen and lung between PAO1 group and 螖 plcH group. The white blood cell count in BALF in PBS group was significantly lower than that in other two groups (P 0.05). P-V curve and decreased delayed inspiratory limb evaluation showed that the pulmonary pathophysiology was very poor in the PAO1 group. Compared with the PBS group, the pulmonary physiological function of the 螖 plcH group was significantly abnormal. Conclusion Pseudomonas aeruginosa plcH may affect pulmonary function by acting on pulmonary surfactant.
