【摘要】：目的對(duì)2例微柱凝膠卡漏檢B_w03亞型標(biāo)本血型血清學(xué)特點(diǎn)及其分子遺傳背景進(jìn)行系統(tǒng)分析,以期為臨床安全輸血提供幫助。方法分別應(yīng)用試管法和微柱凝膠卡法進(jìn)行血型正反定型;應(yīng)用PCR-SSP方法對(duì)ABO血型基因進(jìn)行初步分型;并進(jìn)一步應(yīng)用直接測(cè)序和單體型序列分析法對(duì)ABO基因第6、7外顯子進(jìn)行分析以確定其基因型。結(jié)果標(biāo)本1:試管法結(jié)果疑似B亞型,凝膠卡結(jié)果正定為O型反定為B型,基因檢測(cè)結(jié)果為B_w03。標(biāo)本2:試管法結(jié)果疑似AB亞型,凝膠卡結(jié)果正定為A型反定為AB型,基因檢測(cè)結(jié)果為AB_w03。結(jié)論微柱凝膠卡能漏檢B_w03亞型,應(yīng)采用多種方法進(jìn)行血型鑒定以減少臨床血型鑒定錯(cuò)誤及相關(guān)輸血反應(yīng)的發(fā)生。
[Abstract]:Objective to systematically analyze the blood group serological characteristics and molecular genetic background of two microcolumn gel card missed Bw03 subtype specimens. Methods Blood group positive and negative typing were performed by using test tube method and microcolumn gel card method respectively. The genotyping of ABO blood group gene was carried out by PCR-SSP method. Furthermore, direct sequencing and haplotype sequence analysis were used to analyze exon 6 of ABO gene to determine its genotype. The results of gel card were positively determined as O type and B type, and the result of gene detection was B / W 03. Specimen 2: the test tube method showed that AB subtype was suspected, and the positive definite result of gel card was A type instead of AB type. Conclusion Microcolumn gel card can not detect Bw03 subtype, and blood type identification should be carried out by various methods to reduce the occurrence of clinical blood group identification error and related blood transfusion reaction.
